RNA would most likely be found in organism with high cytoplasmic to nuclear
ratio. Yeast is a good example of this tissue. Different kind
of RNA such as mRNA, tRNA, rRNA may require different method. From
yeaast, RNA requires incubating it with alkaline solution to extract the
nucleic acid and proteins that are HO soluble. This would inactivate nucleasis
that would interfere with the proceeding reaction. After employing this
method, RNA is obtained by acid extraction at pH 4.5 and then precipitated
by the addition of acidified alcohol. Lipids may be an interfering substance
and this should be removed by washings with alcohol or ether or other organic
solutions.
